After being stimulated, cardiomyocytes produce 134 amino acid B-type natriuretic peptide precursor (pre-proBNP), and then form a 108-amino acid BNP precursor(proBNP), which is cleaved to 76 amino acid, NT-proBNP without biological activity and active B-type natriuretic peptide(BNP) containing 32 amino acids.
Although NT-proBNP and BNP have the same biological source, the biological effects and clinical significance are not completely the same.
The concentrations of N-terminal pro atrial natriuretic peptide (ANP) in human serum, plasma and whole blood were measured in vitro.
NT-proBNP is used for early diagnosis, differential diagnosis, treatment guidance and prognosis judgment of heart failure.
The common detection methods are chemiluminescence immunoassay, immunofluorescence chromatography and so on.
The reagent can form a double antibody sandwich complex with NT-proBNP and detect it with fluorescence detector, so as to realize the quantitative determination of NT-proBNP concentration in serum, plasma and whole blood. After the sample is mixed with the buffer solution evenly, it is dropped into the sample hole of the detection card. The sample reacts with the fluorescent labeled mouse anti-human NT-proBNP monoclonal antibody coated on the sample pad to form an antigen antibody complex, which is chromatographed onto the nitrocellulose membrane, and is captured by specific immune reaction with the coated mouse anti-human NT-proBNP monoclonal antibody.
The amount of captured fluorescent complex was positively correlated with the concentration of NT-proBNP in the sample. The fluorescence intensity was detected and converted into NT-proBNP concentration according to the calibration curve calibrated on the ID chip or test card.
【Storage conditions and validity】
The test card and buffer can be stored at 2 ℃～ 30 ℃ for 24 months. After the buffer solution was opened, it could be kept stable for 2 hours at 2 ℃～ 30 ℃. The test card should be used within 2 hours after opening. Transportation and storage should be carried out at 2 ℃ ~ 30 ℃.
